PositionCerebral amyloid angiopathy (CAA) is a common brain disorder, characterized by deployment of amyloid-β (Aβ) in arteriols and capillaries in the brain. Caa is a major cause of brain bleeding, but also contributes to dementia, including Alzheimer's disease. At Alzheimer Patients, CAA forms the substrate for potentially serious side effects ('Aria') of anti-amyloid immunotherapics. We are looking for the PhD student in the course of the Traffic ('Translational Framework for Innovation' in Cerebral Amyloid Angiopathy 'project. The Traffic Project is designed by an international consortium of 7 research groups from the Netherlands, Germany, Spain and the United States.

As a PhD student, you enter the following work / tasks:

Ondanks de hoge prevalentie en klinische impact bestaan er voor aandoeningen binnen het CAA-spectrum onvoldoende sensitieve diagnostische methoden. De huidige diagnostische criteria op basis van MRI hebben beperkte sensitiviteit, vooral in een vroeg stadium en bij de inflammatoire subtypen. Daarnaast ontbreekt in de huidige criteria integratie met moleculaire biomarkers die de onderliggende biologie van de cerebrale vaten weerspiegelen.
Het doel van het internationale TRAFFIC consortium netwerk is om deze onderliggende moleculaire processen in kaart te brengen en te vertalen naar klinisch bruikbare biomarkers, waarmee de kloof tussen mechanistisch onderzoek en diagnostiek op patiëntniveau wordt overbrugd. In het consortium wordt er gewerkt vanuit experimentele diermodellen, hersenweefsel en moderne analyse technieken om biomarkers voor het CAA spectrum in kaart te brengen. Biomarkers worden getoetst in grote klinische cohorten op hun diagnostische waarde. Op basis hiervan worden nieuwe diagnostische modellen gecreëerd en vertaald in klinische richtlijnen. De huidige vacature beschrijft het onderzoek naar de toepassing van nieuwe biomarkers in mensen met CAA en vergelijkbare aandoeningen. Binnen het consortium vormen alle jonge onderzoekers een netwerk van beginnende onderzoekers (‘Leducq CAA Academy’) waarbinnen een breed programma van training aangeboden wordt.
Wij zijn op zoek naar de promovendus die onderzoek verrichten in het kader van het TRAFFIC (‘TRAnslational Framework For Innovation in Cerebral amyloid angiopathy’) project. Het TRAFFIC project wordt vormgegeven door een internationaal consortium van 7 onderzoeksgroepen uit Nederland, Duitsland, Spanje en de Verenigde Staten.

Als promovendus voer je de volgende werkzaamheden/taken uit:

- Rekruteren van patiënten voor de TRAFFIC studie, na uitvoerige training zelfstandig uitvoeren van lumbaal puncties, en neuropsychologisch onderzoek.
- Dataverzameling en analyse van klinische, neuroimaging en biochemische data.
- Inrichten en beheren van een data management systeem.
- Organiseren en coördineren van een internationale samenwerking waarin zo veel mogelijk biomarker informatie van patiënten met CAA verzameld wordt. Deze data integreer je en analyseer je, met als uiteindelijk doel om deze informatie te kunnen toevoegen aan de diagnostische criteria voor CAA.
- Nauwe samenwerking met een multidisciplinair projectteam, inclusief een nog aan te stellen tweede promovendus met een profiel van biomedisch wetenschapper binnen het TRAFFIC project, en andere promovendi die binnen de andere TRAFFIC instituten werkzaam zijn aan dit project.
- Samenwerken met internationale partners; mede-vormgeven aan de samenwerking en training van een groep jonge onderzoekers binnen het consortium.
- Actieve deelname aan consortium bijeenkomsten (live en online).
- Presentatie van de bevindingen in Engelstalige en Nederlandstalige (wetenschappelijke) publicaties, nieuwsbrieven voor deelnemers, en op (inter)nationale congressen.
