Dit betekent dat verkeerd gevouwen Aß-eiwitten als ‘seeds’ kunnen werken die andere Aß-eiwitten aanzetten om dezelfde afwijkende structuur aan te nemen. Als promovendus onderzoek je of Aß40 in het bloed van patiënten met CAA prionachtige eigenschappen heeft. Je werkt daarbij op het grensvlak van fysische chemie, eiwitbiologie, celbiologie en klinische neurowetenschappen. Het onderzoek maakt deel uit van het Vici-project van prof. dr. Marieke Wermer en wordt uitgevoerd in nauwe samenwerking tussen het UMCG en Wageningen University & Research. Je promotietraject start in Wageningen, waar je onder begeleiding van dr. Simo Ruggeri van de afdeling Physical Chemistry and Soft Matter onderzoek doet naar de structurele eigenschappen van Aß40. Vervolgens verplaatst het zwaartepunt zich naar het UMCG, waar je onder begeleiding van dr. Mark Hipp van de afdeling BMS de cellulaire seeding-experimenten uitvoert. Globaal zal je traject er als volgt uitzien:
1. Je brengt amyloid-beta uit bloed op nanoschaal in kaart (WUR). Je onderzoekt Aß40 afkomstig uit plasma van patiënten met sporadische CAA, erfelijke D-CAA en iatrogene CAA en vergelijkt dit met materiaal van patiënten met de ziekte van Alzheimer en gezonde controles. Met onder andere Thioflavin T (ThT)-assays bestudeer je de vorming en kinetiek van amyloidfibrillen.
2. Je karakteriseert de structuur van Aß40 met geavanceerde AFM-IR nanospectroscopie (WUR). Met atomic force microscopy gecombineerd met infraroodspectroscopie (AFM-IR) onderzoek je verschillen in morfologie en moleculaire structuur van Aß40 tussen de verschillende vormen van CAA en controles. Hiermee ontwikkel je als het ware een nanoscale fingerprint van amyloid-beta in menselijk bloed.
3. Je onderzoekt of bloed-afgeleid Aß40 nieuwe amyloidaggregatie kan ‘seeden’ (UMCG). Je ontwikkelt en gebruikt cellulaire Aß40-biosensoren. Met fluorescentiemicroscopie en FRET-gebaseerde technieken onderzoek je of Aß40 uit patiëntenplasma in levende cellen nieuwe aggregatie kan induceren en zichzelf kan voortplanten.
4. Je koppelt de structurele eigenschappen van Aß40 aan de biologische effecten ervan (WUR en UMCG). Je isoleert Aß-aggregaten uit cellen waarin seeding heeft plaatsgevonden en karakteriseert deze opnieuw met onder andere ThT en AFM-IR. Zo onderzoek je of specifieke structurele kenmerken van bloed-afgeleid Aß40 samenhangen met de mate waarin het in staat is nieuwe amyloidaggregatie te induceren.
5. Je onderzoekt de cellulaire gevolgen van Aß40-seeding (UMCG). Met verschillende cellulaire stress-, proteostase- en inflammatoire reporters onderzoek je of seeding leidt tot verstoring van de eiwithuishouding, cellulaire stress en inflammatoire activatie. Daarnaast onderzoek je mogelijke verschillen tussen de verschillende CAA-subtypen en exploreer je sekseverschillen in morfologie en seedingcapaciteit.

Je maakt deel uit van een multidisciplinair onderzoeksteam van neurologen, celbiologen, fysisch chemici en onderzoekers op het gebied van amyloid en eiwitaggregatie. Samen met andere promovendi en een postdoc binnen het Vici-project werk je aan de overkoepelende vraag of CAA mede kan ontstaan door overdracht van amyloid-beta via het bloed.
