Subject of Thesis

Antibioresistance (ABR) is a global threat to human, animal and environmental health. Limiting its tanks
the environment requires a better understanding of its pathways of dissemination. If wastewater and surface water are well studied, the role
soil remains unexplored. Soils are however major reservoirs of antibiotic resistant bacteria (BRAs)
antibiotic resistance gene carriers (GRAs). In this telluric medium, protists (micro-eucaryote), particularly the
amibes, play an important role in the regulation of bacterial populations through their predation activity. However, bacteria
among which opportunistic pathogens, including BRAs, resist amebian lysis after their phagocytosis and persist then
in the ambian microbiota. Amoeba could then play a key role as vectors of resistant bacteria(genes)
antibiotics (BRAs/GRAs). Through an integrated interdisciplinary approach "One Health", the thesis proposes to study the influence of
soil properties on the survival and mobility of amoeba, BRAs and GRAs (molecular markings). Innovative coupling
experiment/modelling (reactors, soil cores, X tomography, hydrobiogeochemical model) will quantify their transport
in soil and predict the risk of release of AMR.

L’antibiorésistance (ABR) est une menace globale touchant la santé humaine, animale et environnementale. Limiter ses réservoirs
environnementaux nécessite de mieux comprendre ses voies de dissémination. Si les eaux usées et de surface sont bien étudiées, le rôle
des sols reste encore peu exploré. Les sols sont pourtant des réservoirs majeurs de bactéries résistantes aux antibiotiques (BRAs)
porteuses de gènes de résistances aux antibiotiques (GRAs). Dans ce milieu tellurique, les protistes (micro-eucaryote), notamment les
amibes, jouent un rôle important dans la régulation des populations bactériennes via leur activité de prédation. Cependant, des bactéries
parmi lesquelles des pathogènes opportunistes dont des BRAs, résistent à la lyse amibienne après leur phagocytose et persistent alors
dans le microbiote amibien. Les amibes pourraient alors jouer un rôle clé comme vecteurs de bactéries(gènes) résistantes aux
antibiotiques (BRAs/GRAs). Par une approche interdisciplinaire intégrée «One Health», la thèse propose d’étudier l’influence des
propriétés des sols sur la survie et la mobilité des amibes, des BRAs et GRAs (marquages moléculaires). Un couplage innovant
expérience/modélisation (réacteurs, carottes de sol, tomographie X, modèle hydrobiogéochimique) permettra de quantifier leur transport
dans le sol et prédire le risque de dissémination de l’AMR.
