Summary: The research fellow will develop the experimental plan under the supervision of the tutor and a senior postdoc. Starting with iPSC lines previously derived from patients’ fibroblasts, the researcher will be responsible for differentiating them into 2D musculoskeletal stem cells and for generating 3D organoid models. To characterise the inflammatory profile, molecular genetics and transcriptomics experiments will be conducted under basal conditions and following LPS-induced stress. For mitochondrial dysfunction, the system will be stressed instead with galactose and similarly analysed. Once a reliable readout has been identified from the stem cell characterisation experiments, a panel of FDA/EMA-approved drugs will be tested to identify an effective therapy. In addition, the researcher will conduct experiments to develop gene therapy using genetic engineering techniques such as CRISPR/Cas9.

Descrizione sintetica: Il titolare dell’incarico di ricerca svilupperà il piano sperimentale sotto la supervisione del tutor e di un senior postdoc. Partendo da linee iPSC precedentemente derivate da fibroblasti dei pazienti, il ricercatore sarà responsabile della differenziazione in cellule staminali muscolo-scheletriche 2D e della generazione da queste di modelli 3D di organoidi. Per la caratterizzazione del pattern infiammatorio verranno condotti esperimenti di genetica molecolare e trascrittomica in condizioni basali e dopo stress con LPS. Per la disfunzione mitocondriale, invece, il sistema verrà stressato con galattosio e analizzato similmente. Individuato un readout affidabile dagli esperimenti di caratterizzazione delle cellule staminali, verrà testata una batteria di farmaci approvati dalla FDA/EMA per individuare una terapia efficace. Inoltre il ricercatore condurrà esperimenti per lo sviluppo di terapia genica con tecniche di ingegneria genetica come CRISPR/Cas9.
