Subject of Thesis

Macrophages are professional phagocytes of the immune system. One of their key functions for both tissue homeostasis and pathogen destruction is to ingest and digest particles. This process called phagocytosis involves a dynamic reorganisation of the cytoskeleton d的actin and the generation of mechanical forces.
When particles are too large to be completely ingested, macrophages can also extract one by one small portion of the target particle, a phenomenon then called trogocytosis. Trogocytosis plays a variety of crucial roles ranging from regulation of the hematopoietic niche to modulation of immune responses. In the context of cancer, this process is used in particular by macrophages to ingest piece by piece of opsonized tumour cells.
During phagocytosis, the macrophage exerts mechanical forces on its target, generated by the polymerization of myosin and the contractility of myosin. If the role of these cytoskeletal motors is well characterized in phagocytosis, the molecular mechanisms and forces involved in trogocytosis remain largely unexplored to date, and we hypothesize that mechanical forces are crucial for effective trogocytosis.
The objective of this project is therefore to characterize the structure and function of cytoskeleton and mechanical forces in the trogocytosis process carried out by macrophages. To do this, experimental models of trogocytosis involving tumour cells and human and murine macrophages will be put in place, and the trogocytosis will be quantified via samples fixed via real-time microscopy approaches. A specific marking strategy will be developed to discriminate between trogocytosis and phagocytosis events. Once these approaches are optimized, pharmacological inhibitors and gene depletion approaches by CRISPR-Cas9 targeting the macrophage cytoskeleton will be used to assess the impact of its inhibition on the efficacy and dynamics of the trogocytosis process, and thus begin to better understand the functioning of this key yet so little detailed process.

Ce projet a donc pour objectif de caractériser la structure et fonction du cytosquelette et des forces mécaniques dans le processus de trogocytose réalisé par les macrophages. Pour ce faire, des modèles expérimentaux de trogocytose impliquant des cellules tumorales et des macrophages humains et murins seront mis en place, et la trogocytose sera quantifiée via des échantillons fixés via des approches de microscopie en temps réel. Une stratégie de marquage spécifique sera développée afin de discriminer les événements de trogocytose de ceux de phagocytose. Une fois ces approches optimisées, des inhibiteurs pharmacologiques et des approches de déplétions de gènes par CRISPR-Cas9 ciblant le cytosquelette des macrophages seront utilisés afin d’évaluer l’impact de son inhibition sur l’efficacité et les dynamiques du processus de trogocytose, et ainsi commencer à mieux comprendre le fonctionnement de ce processus clé et pourtant si peu détaillé.
