The diffusion and diffraction methods of time-resolved X-rays have revolutionized structural biology, highlighting the structural dynamics that accompany essential biological functions. The doctoral student will use the static and time-resolved diffusion of X-rays in solution to track the structural changes of photowitches based on azobenzene-G-quadruplex (AzO-G4) DNA after light excitation, thus revealing the volume variations induced by photoisomerization of the azobenzene and providing insight into the dynamics and structural hierarchy of conformational changes in the AzO-G4.

Located on the Scientific Polygon of Grenoble (ESRF/ILL/EMBL/IBS), the Institute of Structural Biology (IBS) is a national and international player in the field of integrated structural biology. At the same time a research centre, a technical platform, a site of reception and scientific training, the IBS has as its purpose the development of research in structural biology, a field of research which is crucial for the understanding of fundamental biological mechanisms. It relies on 12 state-of-the-art platforms.
As a joint research unit (CEA-CNRS-UGA) the IBS is composed of 20 research groups, each of which proposes a multidisciplinary approach, at the boundaries of biology, physics and chemistry, consistent with 3 research axes. Nearly 320 people work there: researchers, PhD students, engineers and technicians in a multicultural and international environment.

Activities:

Les méthodes de diffusion et de diffraction des rayons X résolues en temps ont révolutionné la biologie structurale, en mettant en lumière la dynamique structurale qui accompagne des fonctions biologiques essentielles. Le doctorant utilisera la diffusion statique et résolue en temps des rayons X en solution pour suivre les changements structuraux des photoswitches à base d’ADN azobenzène-G-quadruplex (AzO-G4) après excitation par la lumière, révélant ainsi les variations de volume induites par la photoisomérisation de l’azobenzène et apportant un éclairage sur la dynamique et la hiérarchie structurelle des changements conformationnels de l’AzO-G4.
