This thesis project will focus on the question of mechanicals by the filaments of the actin and vimentin cytoskeletians, focusing on the family of proteins at LIM domain, some being already known to recognize the mechanical state of the filaments. This is an experienced project based on in vitro reconstitution experiments within a microfluid chamber and using purified and fluorescent proteins. Ce projet de thèse s’intéressera à la question de la mécanosensation par les filaments des cytosquelettes d’actine et de vimentine, en se focalisant sur la famille des protéines à domaine LIM, certaines étant déjà connues pour reconnaître l’état mécanique des filaments. Il s’agit d’un projet expérimenté basé sur des expériences de reconstitution in vitro au sein d’une chambre microfluidique et utilisant des protéines purifiées et marquées en fluorescence. Le ou la candidate devra quantifier la réponse mécanique de filaments uniques et la dynamique d’assemblage de certaines protéines à domaine LIM. L’objectif final sera de proposer un modèle de reconnaissance de l’état mécanique des filaments et de la formation des assemblages de protéines à domaines LIM, afin d’éclairer les mécanismes de mécanotransduction dans les cellules.
