Forskarutbildningsämne:Biotechnology
At the Department of Protein Science, we develop predictable and robust techniques for production, purification and detection of proteins. As building blocks for new molecular features, we use small, structurally well-characterized protein domains. The doctoral project is based on our newly developed calcium-regulated affinity proteins (Caras), which binds to its target proteins in the presence of calcium and releases them at low calcium concentrations. By combining proteinengineering with biophysical characterization, structural biology, calculation modeling and machine learning, you will examine how the amino acid sequence and the surroundings ratios control this behavior. The project unites the expertise that has been developed in Professor Sophia Hober's research group at the Royal Institute of Technology (KTH) and Professor Andreas Hougaard Laustsen research group at Denmark Technical University (DTU). The goal is to clarify the principles behind environmentally regulated protein function and translate them into design principles to develop a new generation smarter and more precise therapeutic proteins.
Tutorial:Sophia Hober is proposed to supervise the doctoral student. Decisions are taken on admission

Information about admission and employment
Only the one who has been adopted to postgraduate education may be employed as a doctoral student. The total employment period may not be longer than that corresponds to education at full-time postgraduate for four years. An employee doctoral student can to a limited extent (at most 20 %) perform certain tasks within eg. education and administration. A new employment as a doctoral student applies for a maximum of one year, the employment may then be renewed by a maximum of two years at a time. In studies to be terminated with a licentiate degree, the total employment time may not be longer than that corresponds to training at a full-time postgraduate for two years.
As a doctoral student, you get part of KTH's employment benefits and has a monthly salary according to KTH's doctoral salary agreement. Read more about research studies at KTH | KTH | Sweden.

Project description
Third-cycle subject: Biotechnology
At the Department of Protein Technology, we develop predictable and robust technologies for protein production, purification, and detection using small, structurally well-characterized protein domains as building blocks for new molecular functions. This PhD project builds on our recently developed calcium-regulated affinity binders (CaRAs), which bind their targets in the presence of calcium and release them at low calcium concentrations. By combining protein and protein engineering with biophysical characterization, structural biology, computational modelling, and machine learning, you will investigate how amino acid sequence and environmental conditions govern this behaviour. The project brings together the expertise built in Professor Sophia Hober´s group at KTH Royal Institute of Technology and Professor Andreas Hougaard Laustsen’s gruop at the Technical University of Denmark. The project aims to uncover the principles governing environmentally regulated protein function and translate them into design rules for a new generation of smarter and more precise therapeutic proteins.
