Hyperaccumulation of vanadium in sponge holobionts: bacterial contribution and physiological role.

Your Working Environment

The hyperaccumulation of the metal element vanadium (V) has recently been shown in two homoscleromorphic sponges. The objectives of the thesis are 1) to determine whether bacterial communities are themselves affected by hyperaccumulation and/or involved in the process (metabolic pathways), 2) what is the physiological role of this hyperaccumulation, 3) to participate in the study of molecular mechanisms? and 4) assess the possibility of using these holobionts to sequester V for bioremediation or recycling. The thesis project is part of a larger project funded by ANR involving four laboratories; the study implemented will therefore be based on an integrative approach.
MISSIONS
The PhD student will have to be involved in the preparation of biological samples and experimentally in the various techniques used to meet the previous objectives (bioinformatics, physiology, chemistry, cell biology: an integrative approach from molecule to ecosystem). It will have to ensure a bibliographical monitoring and present its results to the consortium on a regular basis. He/she will have to co-author articles from his/her work, present his/her results at congresses and write his/her thesis manuscript.
ACTIVITIES
- Separate bacteria from hyperaccumulative sponge cells by flow cytometry and then evaluate by ICPMS if symbiotes also accumulate vanadium
- Compare by metabarcoding if bacterial communities are different between close hyperaccumulative and non-hyperaccumulative species, if necessary determine possible bacterial families likely to be involved in this mechanism.
- Participate in characterization of the expression of candidate genes or in the location of proteins potentially in the hyperaccumulation mechanism
- Compare V levels between individuals exposed or not exposed to V
- Compare tolerance to oxic stress of accumulating and non-accumulative species
- Compare chemical compounds emitted by accumulating and non-accumulative species

L’hyper accumulation de l’élément métallique vanadium (V) a été montrée récemment chez deux éponges homoscléromorphes. Les objectifs de la thèse visent 1) à déterminer si les communautés bactériennes sont concernées elles-mêmes par l’hyper accumulation et/ou impliquées dans le processus (voies métaboliques), 2) quel est le rôle physiologique de cette hyper-accumulation, 3) participer à l’étude des mécanismes moléculaires ? impliqués et 4) évaluer la possibilité d’utiliser ces holobiontes pour séquestrer du V à des fins de biorémédiation ou recyclage. Le projet de thèse s’inscrit dans un projet plus large financé par l’ANR qui implique quatre laboratoires ; l’étude mise en œuvre s’appuiera donc sur une approche intégrative.
MISSIONS
Le doctorant devra s’impliquer dans la préparation des échantillons biologiques et expérimentalement dans les différentes techniques mises en œuvre pour répondre aux objectifs précédents (bioinformatique, physiologie, chimie, biologie cellulaire : approche intégrative de la molécule à l’écosystème). Il devra assurer une veille bibliographique et présenter régulièrement à l’oral ses résultats devant le consortium. Il/elle devra rédiger en collaboration les articles issus de ses travaux, présenter ses résultats lors de congrès et rédiger son manuscrit de thèse.
ACTIVITÉS
- Séparer les bactéries des cellules d’éponges hyper accumulatrices par cytométrie en flux puis évaluer par ICPMS si les symbiotes accumulent également le vanadium
- Comparer par metabarcoding si les communautés bactériennes sont différentes entre espèces proches hyper accumulatrices et non hyper accumulatrices, le cas échéant déterminer d’éventuelles familles bactériennes susceptibles d’être impliquées dans ce mécanisme.
- Participer à la caractérisation de l’expression des gènes candidats ou à la localisation des protéines potentiellement dans le mécanisme d’hyperaccumulation
- Comparer les teneurs de V entre individus exposés ou non au V
- Comparer la tolérance au stress oxique des espèces accumulatrices et non accumulatrices
- Comparer les composés chimiques émis par les espèces accumulatrices et non accumulatrices
