Subject of Thesis
a resistance to drugs and toxicity underline the urgent need for new strategies to discover and design bioactive natural products. The purpose of this project is to determine the molecular mechanism of pyrmidin cycle assemblage during bleomycin biosynthesis, a glycopeptide antibiotic, and an anticancer drug, through a non-ribosomal peptide (NRPS) synthesis chain. Pyrimid fragments are essential to the activity of many drugs, while the modular architecture of NRPS makes them interesting platforms for bioengineering. Our project will combine biochemistry, structural biology and chemical synthesis to define the catalytic mechanisms, structures and protein-protein interactions responsible for the formation of NRPS mediated pyrimidines. By clarifying how the specialized areas of NRPS cooperate in building complex heterocycles, we will highlight the molecular foundations of their unusual chemistry and the interaction interfaces that govern biosynthetic efficiency. These results will establish a framework for the rational engineering of NRPS to produce new natural products containing pyrimidines with improved bio-activity, thereby promoting sustainable approaches to drug resistance and toxicity.

Ce projet vise à déterminer le mécanisme moléculaire de l'assemblage du cycle pyrimidinique au cours de la biosynthèse de la bléomycine, un antibiotique glycopeptidique et un médicament anticancéreux, par une chaîne de synthèse de peptides non ribosomaux (NRPS). Les fragments pyrimidiques sont essentiels à l’activité de nombreux médicaments, tandis que l’architecture modulaire des NRPS en fait des plateformes intéressantes pour la bio-ingénierie. Notre projet associera la biochimie, la biologie structurale et la synthèse chimique afin de définir les mécanismes catalytiques, les structures et les interactions protéine-protéine responsables de la formation des pyrimidines médiée par les NRPS. En élucidant la manière dont les domaines spécialisés des NRPS coopèrent pour construire des hétérocycles complexes, nous mettrons en lumière les fondements moléculaires de leur chimie inhabituelle et les interfaces d’interaction qui régissent l’efficacité biosynthétique. Ces résultats établiront un cadre pour l’ingénierie rationnelle des NRPS afin de produire de nouveaux produits naturels contenant des pyrimidines dotés de bioactivités améliorées, favorisant ainsi des approches durables pour lutter contre la résistance aux médicaments et la toxicité.
